The pilot first aired as a special presentation on June 19, 2005, and the series later started officially airing from October 23, 2005 until ending on December 17, 2006, with a total of 20 episodes over the course of 2 seasons. A special, entitled "Spider-Man Special", aired on November 6, 2005 and a stand-alone webisode, entitled "Enter the Sandmouse", was later released online on May 16, 2007.

Rectangular Businessman executes Rhoda by telepathically slicing his head in half, revealing a fire-breathing snake inside. Fitz takes down The New Guy with a rocket-powered skateboard and helps Skillet and the rest escape the shack. Skillet discovers an invitation to a fancy party Shark is holding, and Fitz decides to crash it. Shark and Rectangular Businessman get fed up with the Eye and decide to punish him. Rectangular Businessman makes Pronto enter Fitz's house to free the Hand from his icebox. The Hand subsequently cuts off the Eye's leg with a pair of shears. At the party, Fitz is sucked into a passageway behind a bookshelf. As Skillet is shot with a dart and Eye collapses in a puddle of blood, Shark watches from his control room and laughs evilly.

Fitz, Skillet, Cyber-Green-Sweatered-Woman, and Peanut Cop travel through the ventilation shaft and shoot several tie-bots inside. Peanut Cop throws a grenade and blasts his fireman's helmet off. Fitz, Skillet, and Cyber-Green-Sweatered-Woman burst out the end of the ventilation shaft, which opens into Shark's observation room. Together, they de-shackle the Eye, destroy the evil Eye, shoot Shark into bloody pieces, and set Golden Joe free. After hearing some faint music emanating from Shark's remains, Fitz digs inside of them and pulls out the New Guy, who has been partially digested. Fitz pulls a lever on Shark's video monitoring console, which opens a room containing a metal-plated, shark-shaped aircraft. The Eye says that he's afraid of flying, so he decides to walk to the river and carry the New Guy on top of him.

The plot revolves around a bizarre conspiracy which is constantly building on itself and taking seemingly nonsensical twists and turns. At times the narrative becomes obfuscated or nonlinear, but it simultaneously reveals connections that hint at an objective story. Clues and secret messages are hidden throughout each episode, including the credits. Creator Matt Maiellaro has indicated that the development of 12 oz. Mouse was influenced in part by surrealistic films and the Theatre of the Absurd, such as those by director David Lynch.

The series concluded its initial run on Adult Swim on December 18th, 2006 with its twentieth episode, leaving the plot unresolved. In February 2007, production began on additional episodes for internet distribution on Adult Swim Video.[3] The first and only online episode premiered May 16th, 2007.

Maiellaro spent three weeks working with Ingkatanuwat on putting together the set for the opening title sequence. The set was filmed with a motion control camera, and was inserted with miniature explosives and smoke bombs for special effect. Nine Pound Hammer composed the opening theme song; Maiellaro sought for a song representing the "carefree" lifestyle of Mouse who "does things like drive drunk, film porno and shoot guns."[2] Maiellaro, who plays the electric guitar in his free time, also composed the song "F-Off", featured in the first episode, which he wrote while working on Space Ghost Coast to Coast.[2] Swedish heavy metal band, Amaranthe performed the credits theme song for the 2018 special, "Invictus."

On October 14th, 2018, a half-hour special titled "Invictus" premiered on TV after having been released online two days earlier. Simultaneously, Adult Swim announced the series would be returning for a third season.

Liquor runs the liquor store where Fitz frequently stops. His temperament can best be described as "resigned." He seems to be Fitz's ally and is defiant to anyone of authority, such as Shark and Rectangular Businessman. Liquor seems to know about the secret nature of the world: in "Surgery Circus" he uses a button to freeze Shark's illusions, he reveals a duplicate of Eye that was hidden inside his store, and in "Eighteen" he discovers and communicates with the Hand while it's inside Producer Man's brain. He also knows about Roostre's connection with the Corn Droid and recognizes Muff.

After "Adventure Mouse", Golden Joe remains allied with Fitz. He is later captured by Shark until Fitz and his friends free him in "Farewell". He remains with Fitz and Skillet until the opening webisode of the "third season", where he is abducted by a Hawk.

A bee who is an exterminator that Fitz hires when he finds Spider in his house back in the real world. Buzby chases Spider out with a chaingun, and then saves Fitz from being blown up by Shark. It is later revealed though that Buzby was working with Shark all along, wanting to bring Fitz back to the alternate dimension to free Shark and Rectangular Businessman. 2ff7e9595c
